What Is DSIP? A Research Overview of Delta Sleep-Inducing Peptide

Dsip

DSIP, or delta sleep-inducing peptide, is a nonapeptide first isolated in sleep research in the 1970s. This overview covers its structure, discovery, research history and regulatory status. It is a scientific summary, not guidance on use.

Structure and origin

DSIP has the sequence Trp-Ala-Gly-Gly-Asp-Ala-Ser-Gly-Glu (WAGGDASGE), nine amino acids; its international nonproprietary name is emideltide. It was isolated in 1977 from the cerebral venous blood of rabbits by Monnier, Schoenenberger and colleagues in Switzerland (Monnier et al., Experientia, PMID 862769). No gene, precursor protein or receptor for DSIP has been identified. See the DSIP product page.

How DSIP has been studied

  • Early clinical work in insomnia (Schneider-Helmert et al., Lancet, 1981, PMID 6112579).
  • A double-blind study in chronic insomnia (Bes et al., Neuropsychobiology, 1992, PMID 1299794).
  • Stress-hormone (corticosterone) release in rats (Graf et al., Neuroendocrinology, 1985, PMID 2995861).
  • Opioid withdrawal (Soyka et al., Am J Psychiatry, 1997, PMID 9137140).

A 2006 review discusses why DSIP’s biology remains unresolved (Kovalzon and Strekalova, J Neurochem, PMID 16539679). Human results have been mixed, and the FDA described the evidence for chronic insomnia as insufficient in 2026.

Regulatory status

  • DSIP is not approved by the U.S. Food and Drug Administration for any use, and the FDA did not find it registered as a drug in any country it searched.
  • In July 2026 the FDA Pharmacy Compounding Advisory Committee voted 6–7 against recommending DSIP for the compounding list.
  • DSIP is not named on the WADA prohibited list; non-approved substances may fall under class S0. Check Global DRO.

Laboratories should confirm the current status in their jurisdiction before beginning work.

Quality considerations for researchers

Results depend on the material used, so identity and purity data matter. Check that each batch comes with a certificate of analysis showing mass-spectrometry identity and HPLC purity, and that the batch number matches your vial. Store lyophilized material as described in our storage guide.
